The Effect of gp130 on Osteoclastic Differentiation and Activation

Abstract
Signals through gp130 induce diverse biological effects in immune, hematopoietic, and nervous systems. Substantial evidence also indicates that the IL-6 related cytokines have profound effects on the bone metabolism by regulating both osteoclastic and osteoblastic development and function. However, their mechanisms were not well elucidated. To understand the effects of gp130-mediated signals on osteoclastic differentiation and activation, we investigated the developing skeleton of gp130 deficient mice by skeletal preparation with histological examination focusing on tibia through routine H&E and von Kossa staining. The TRAP activity was also detected histochemically. The expression pattern of osteoclast marker genes, TRAP and MMP-9 mRNAs, was evaluated by in situ hybridization. gp130 deficient mice revealed retarded skeletal development with bending of long bones of digits. Histologically they showed poor calcified matrix formation with marked reduction of primary spongiosa and concentration of numerous large round shaped multinucleated TRAP (+) cells along the border of growth plate. The gp130 deficient osteoclasts demonstrated poor development of ruffled border in electron microscopic observation. These osteoclasts expressed higher TRAP mRNA signals compared to wild type osteoclasts whereas MMP-9 expression in osteoclasts located at diaphysis of gp130 deficient mice was reduced compared to that of wild type osteoclasts. These finding suggest that the gp130-mediated signals may important for full activation of osteoclasts rather than osteoclastic differentiation.
