The Effects of C161→T Polymorphisms in Exon 6 of Peroxisome Proliferator-Activated Receptor-γ Gene on Bone Mineral Metabolism and Serum Osteoprotegerin Levels in Perimenopausal Korean Women

Abstract
Background
Peroxisome proliferator-activated receptor (PPAR)γ is mainly known to be involved in adipocyte differentiation. Recent studies revealed the inhibitory role of PPARγ in osteoblastogenesis and suggested the possibility as the candidate gene for osteoporosis. We observed the frequency of C161→T substitution in exon 6 of PPARγ in Korean females and examined the association of different genotypes and the menopausal status with bone turnover markers, bone mineral density (BMD) and serum osteoprotegerin (OPG), which plays an inhibitory role in osteoclastogenesis.
Methods
In 263 healthy Korean women (mean age 51.5±6.8 yrs; range 37~73 yrs), anthrophometric measurements were done and lumbar spine and femoral neck BMD, bone turnover markers, such as alkaline phosphatase (ALP), serum calcium, phosphorus, 24 hrs urine calcium, phosphorus excretion and urine deoxypyridinoline were measured. Serum follicular stimulating hormone (FSH) levels were measured and serum OPG levels were measured with sandwich enzyme-linked immunosorbent assay (ELISA) method. DNAs were extracted from the samples and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was performed and the sequencing of the products was done to confirm the substitution.
Results
Allele frequencies were 0.804 for C allele and 0.196 for T allele, which was in Hardy-Weinberg equilibrium (p=0.94). There were no differences in mean age, body mass index (BMI), BMD and bone turnover markers among different genotypes and subjects with T alleles showed significantly lower serum OPG levels (p=0.003). There were no differences in the prevalences of osteoporosis according to the genotypes. Analyzed according to the menopausal status, only in postmenopausal women, subjects with T alleles had significantly lower serum OPG levels, which showed consistently significant differences after adjustment for age and FSH (p<0.05).
