Effect of TGF-β1 and BMP-2 for the Chondrogenesis of Human Bone Marrow Stem Cells

Abstract
Purpose
The aim of this study was to determine the effect of TGF-β1 or BMP-2 on the chondrogenesis of adult human bone marrow stem cells. Material and Methods: Human bone marrow stem cells were isolated from marrow aspirates of iliac crest, and adherent cell cultures were established. The cells were subsequently exposed to 0, 10, 50, 100 ng/ml of BMP-2 and 1, 5, 10 ng/ml of TGF-β1 in a serum-free medium to make 7 groups. The chondrogenesis of culture products was then assessed in terms of glycosaminoglycan (GAG) production with alcian blue staining and dimethylmethylene blue dye assay, total collagen with sirius red dye assay, and type II collagen with immunocytochemistry. We performed 6 repeated independent experiments.
Results
Treatment with 100 ng/ml BMP-2 significantly increased the formation of GAG and total collagen, but did not increase type II collagen production (p>0.05). In cultures treated with 10 ng/ml TGF-β1, there was a significant increase in the production of GAG, total collagen, and type II collagen (p<0.05).
Conclusion
Our results suggest that 10 ng/ml TGF-β1 has a stimulatory effect on the chondrogenesis of adult human bone marrow stem cells. Further studies must be conducted to growth factors or other elements to stimulate chondrogenesis in vitro or in vivo.
